The next year, PacBio developed a new method to allow multiple sequencing passes on the same DNA molecule. Double stranded DNA templates were ligated to two single stranded adapters, creating what the company called a “SMRTbell template.” Sequencing began at a primer on one of the adaptors and could proceed multiple times per molecule due to circularization, in a process called rolling-circle amplification.
